ABSTRACT

Cardiovascular diseases cause significant morbidity and mortality worldwide, incurring a major public health burden. Gastrodia elata Blume is a traditional Chinese herbal medicine that has been widely used to treat central nervous system and cardiovascular diseases. Gastrodin, as the major active component in Gastrodia elata Blume, can confer protection against cardiovascular diseases. In this review, we summarize the anti-inflammatory actions, anti-cardiac hypertrophy, anti-hypertension, anti-atherosclerosis, and angiogenic effects of gastrodin, as well as its protective effects on vascular cells and against myocardial ischemia-reperfusion injury. The medical potential of gastrodin in diabetes-related cardiovascular diseases is also discussed.

ABSTRACT

Aim To verify the mechanisms underlying the protective effects of the 4-hydroxybenzaldehyde (4-HBd)on cerebral ischemia / reperfusion in vivo and in vitro. Methods The model of focal cerebral ischemi-a / reperfusion in rats was established by the suture-oc-clusion method. After reperfusion,neurological score and cerebral infarction rate were assessed. Meanwhile, after pretreatment with 4-HBd for 24 h,the primary cultured cortical neuron cells were exposed to oxygen glucose deprivation / reoxygenation (OGD/ Rep). The survival rate, MDA, SOD, NO, Bax, Bcl-2 and caspase-3 concentrations were assessed. Results Pre-treatment with 4-HBd significantly decreased neurologi-cal score (P < 0. 05)and cerebral infarction rate (P <0. 01)in the cerebrum after MCAO/ R injury. Further-more,4-HBd significantly increased cell survival rate (P < 0. 05),decreased MDA content (P < 0. 05),in-creased SOD activity (P < 0. 05),decreased NO levels (P < 0. 05),decreased caspase-3 (P < 0. 05),in-creased Bcl-2 (P < 0. 05)but decreased Bax (P <0. 05)levels and therefore increased Bcl-2 / Bax ratio in the cerebrum after OGD/ R injury. Conclusions 4-HBd has significant protective effect on MCAO/ R rats, and the mechanism may be related to decreasing NO levels,Bax,Bcl-2,caspase-3 and reducing neuronal apoptosis.

ABSTRACT

AIM To investigate the anti-neuroinflammation effects of 4-hydroxybenzyl aldehyde (4-HBAL) from Gastrodia elata Blume on acute cerebral ischemic injury in rats and its nechanism of action.METHODS The rat model of acute cerebral ischemic injury was induced by injecting arachidonic acid via intracarotid artery.Brain tissue samples were taken from the animals 3 h after the model of acute cerebral ischemic injury.Tumor necrosis factor-alpha (TNF-α) and interleukin-1 beta (IL-1β) were detected in brain tissue to evaluate the effects of 4-HBAL in vivo.Lipopolysaccharid (LPS)-induced activation of BV-2 microglia cells model was used to explore the anti-neuroinflammation mechanism of 4-HBAL.RESULTS The experimental results showed that 4-HBAL had a significant protective effect on acute cerebral ischemic injury.It could significandy decrease the contents of tumor necrosis factor-alpha (TNF-α) and interleukin-1 beta (IL-1β),and obviously inhibit the production of nitric oxide (NO),prostaglandin E2 (PGE2) and TNF-α in LPS-stimulated BV-2 cell,and increase the production of interleukin-10 (IL-10) and transforming growth factors-β (TGF-β) in BV-2 cell.CONCLUSION The mechanism of 4-HBAL may be related to the suppression of the excessive activation of microglia after cerebral ischemia and the promotion of the transformation of microglia into anti-inflammatory phenotype.

ABSTRACT

Seven new 4-hydroxybenzyl-substituted amino acid derivatives (1-7), together with 11 known compounds, were isolated from an aqueous extract of the rhizomes of Gastrodia elata Blume. Their structures were determined by spectroscopic and chemical methods. Compounds 1-3 are pyroglutamate derivatives containing 4-hydroxybenzyl units at the N atom and 4-7 are the first examples of natural products with the 4-hydroxybenzyl unit linked via a thioether bond to 2-hydroxy-3-mercaptopropanoic acid (4-6) and 2-hydroxy-4-mercaptobutanoic acid (7), which would be biogenetically derived from cysteine and homocysteine, respectively. The structures of 1 and 2 were verified by synthesis, while the absolute configurations of 4, 5 and 7 were assigned using Mosher's method based on the MPA determination rule of Δδ RS values. The known compound 4-(hydroxymethyl)-5-nitrobenzene-1,2-diol (8) exhibited activity against Fe(2+)-cysteine induced rat liver microsomal lipid peroxidation with IC50 values of 9.99×10(-6) mol/L.

ABSTRACT

Gastrodia elata Blume is a well-known kind of natural products used as a folk medicine for thousands of years. However, anti-atopic dermatitis-like effects of G. elata Blume had not been evaluated until now. The aim of the present study is to investigate the protective effects of water extract from the roots of G. elata Blume (GE) on 2, 4-dinitrochlorobenzene-induced atopic dermatitis-like skin lesions using balb/c mice. Combination treatment of GE (at a dose of 12.5 mg/kg body weight by administrated per os + 0.5 mg/cm2 as ointment to apply on ear and dorsal skin) was significantly inhibited spleen weight, ear thickness, levels of serum immunoglobulin E and number of mast cells, compared with that of 2, 4-dinitrochlorobenzene-included groups without GE. Furthermore, combination application by oral administration plus by ointment of GE significantly inhibited the histamine release from rat peritoneal mast cells. These results suggest that combination treatment of oral administration plus ointment form of GE could be helpful as a potentially natural pharmaceutical treatment on atopic-like dermatitis.

ABSTRACT

Objective: To study the chemical constituents of Gastrodia elata. Methods: The compounds were separated and purified by column chromatography and their structures were established by spectroscopic methods. Results: Nine compounds were isolated from the 50% ethanol extract of G. elata, whose structures were determined as 1-furan-2-yl-2-(4-hydroxy-phenyl)-ethane-1, 2-dione (1), 2, 4-bis (4-hydroxybenzl)-phenol (2), 4-(methoxymethyl) phenyl-1-O-β-D-glucopyranoside (3), 1-furan-2-yl-2-(4-hydroxy-phenyl)- ethanone (4), parishin B (5), parishin C (6), {1-[4-(β-D-glucopyranosyl-(1→3)-β-D-glucopyranosyloxy) benzyl], 2-[4-(β-D-glucopyranosyloxy) benzyl]} citrate (7), bis (4-hydroxybenzyl) sulfide (8), and di-(p-hydroxybenzy1) disulfide (9). Conclusion: Compound 1 is a new compound, named gastrofurodione, and compound 9 is isolated from this genus for the first time.

ABSTRACT

Objective: To study the chemical constituents of Gastrodia elata. Methods: The constituents were isolated by various chromatographies, including preparative TLC, silica gel, Sephadex LH-20, and ODS columns, and their structures were elucidated on the basis of their physicochemical properties and spectral data. Results: Eleven compounds were isolated from 70% EtOH fraction eluted from resin column and their structures were identified as gastrodin (1), 4-hydroxybenzyl alcohol (2), 4-hydroxybenzoic acid (3), 4-hydroxybenzaldehyde (4), 4-hydroxybenz-yl methyl ether (5), parishin (6), 4-(methylsulfinylmethyl) phenol (7), dibutyl phthalate (8), dioctyl phthalate (9), 4, 4'-dihydroxydiphenyl methane (10), and grossamide (11). Conclusion: Compound 7 is a new natural product, and compounds 9 and 11 are isolated from the plants of Orchidacee for the first time.

ABSTRACT

The germination rate and longevity of seeds of Gastrodia elata Blume have been observed for 48 weeks using Mycena osmundicola strain H-21, one of fungi stimulating seed germination. Storage condition of post-harvest seeds was observed in the different temperature ranges of -30degrees C, -5degrees C, 5degrees C and 30degrees C for 48 weeks. After storage period of 48 weeks, the germination rate of G. elata was 65.7% at 5degrees C and 71.6% at -5degrees C, respectively. Although the germination rate of G. elata was 77.3% for 11 weeks at 25degrees C, the germination rate had been decreased gradually to 49.3% at 13 weeks, 0.3% at 23 weeks and then 0% at 25 weeks. The germination rate was reached to the level of 10% for 2 weeks at -30degrees C and then decreased to 0%.

ABSTRACT

Aim To study the effect of polysaccharides from Gastrodia elata Blume(GEP) on levels of superoxide dismutase(SOD),malondialdehyde(MDA) and nitric oxide(NO);and the retinal morphology after rat retina ischemia reperfusion injury.Method Retinal ischemia was induced in SD rats by increasing the intraocular pressure to 16 kPa for 60 min via cannulation into the anterior chamber.SD rats were divided into five groups randomly: Small dose GEP group,Middle dose GEP group,Large dose GEP group,Vitamin E group and Negative contral group.At 60 min,24 h and 72 h after ischemia reperfusion,levels of SOD,MDA and NO in rat retinas were measured,and histopathologic changes were observed.Results At 60 min,24 h and 72 h after ischemia reperfusion,levels of SOD in rat retinas in middle,large dose GEP groups were significantly higher than that in negative control group(P

Objective To study the high polar constituents of Gastrodia elata.Methods The constituents were isolated by macroporous resin column chromatography combined with preparative HPLC and identified by spectral analyses and physico-chemical properties comparisons.Results Ten compounds were isolated from H2O and 20% EtOH fractions eluted from resin column and their structures were identified as citric acid monoethyl ester(Ⅰ),uridine(Ⅱ),adenine(Ⅲ),dactylose A(Ⅳ),uracil(Ⅴ),tyrosine(Ⅵ),N2-(p-hydroxybenzyl) guanosine(Ⅶ),1-isoferuloyl-?-D-glucopyranoside(Ⅷ),4-(?-D-glucopyranosyloxy) benzaldehyde(Ⅸ),and p-hydroxyl benzoic acid(Ⅹ).Conclusion Compound Ⅶ is a new compound named gastronucleoside.Other nine compounds are isolated from G.elata for the first time.

ABSTRACT

Objective Using the soaking seedling method to introduce wild Gastrodia elata DNA into potato plantlets and analyze gastrodin of transformed Solanum tuberosum.Methods After the tuber grown up,the solution of gastrodin was extracted from the potatoes which were transformed by wild G.(elata) DNA.The transformed plants were scaned by ultraviolet.PCR was used to analyze GAFP gene by SDS-PAGE.TLC was used to analyze the gastrodin of transformed S.tuberosum.Results(1)The 21 transformed plants had obvious absorption peak at 220 nm in 200 transformed S.tuberosunr.(2)The fragment of wild G.elata GAFP gene was found by PCR from 5 transformed plants.(3)The protein expressions in transgenic S.tuberosum were obviously different from the normal S.tuberosum;a band was detected in transgenic S.tuberosum and wild G.elata,but wasn't detected in normal S.tuberosum.(4)The expression of the gastrodin was detected by TLC from three transformed plants. Conclusion It is feasible to use the soaking seedling method to introduce exogenous DNA into plants.
